Bacterial Expression Vectors

Vectors have a lac promoter and are intended to be used as expression vectors in bacterial systems. Vectores can also be used as a source of different fluorescent protein coding sequences (genes), which can be amplified by PCR.

EF-1 Alpha Promoter Vectors

Human elongation factor-1 alpha (EF-1 alpha) is a constitutive promoter of human origin that can be used to drive ectopic gene expression in various in vitro and in vivo contexts.

  • Living Colors fluorescent protein fusion vectors
  • IRES vectors for coexpression of 2 genes
  • Vectors with highly immunoreactive Myc or HA tags
  • Can be used to confirm Matchmaker interactions in mammalian cells

Fucci Cell Cycle Vectors

Cell cycle reporter vectors allow monitor cell cycle progression in living cells, in real-time, without fixation. They even let you visualize cell shape.

  • Retroviral vectors to track cell cycle phase in living cells
  • Differentiate G1 phase from S/G2/M based on color
  • See the complete morphology of the cell during simultaneous monitoring of cell cycle phase

Fusion protein products

  • Cyan Fluorescent Protein- AmCyan1 Fluorescent Protein
  • Far Red Fluorescent Proteins
  • Green Fluorescent Protein (GFP) Fusions- AcGFP1 & ZsGreen1
  • mOrange Fluorescent Protein
  • Photoswitchable Fluorescent Protein-Dendra2
  • Red Fluorescent Proteins
  • Yellow Fluorescent Proteins-mBanana and ZsYellow1

MicroRNA Expression Vectors

These vectors provide strong miRNA expression and allow you to verify and track that expression with a fluorescent protein marker.

  • Strong coexpression of miRNA and a bright fluorescent protein
  • Clone your miRNA into the 3'UTR of mCherry or ZsGreen1
  • Simultaneously verify miRNA expression and transfection efficiency via fluorescent microscopy

Photoactivatable-mCherry Fluorescent Protein

Living Colors PAmCherry is a photoactivatable fluorescent protein (PAFP) derived from the red fluorescent protein mCherry. PAmCherry is non-fluorescent until it is exposed to 350–400 nm light.

  • Non-fluorescent until activated by a short exposure to 350–400 nm light
  • Excitation maximum: 564 nm
  • Emission maximum: 595 nm

Photoconvertible Fluorescent Proteins-Dendra2

Dendra2 is a photoswitchable (green to red), monomeric fluorescent protein derived from octocoral Dendronephthya sp.

  • Irreversible switching from green to red is activated by UV-violet or blue light
  • Switched (red) form is highly photostable
  • Matures at a wide range of temperatures

Promoter reporters

Promoterless (Lowest Background)

On-demand reporter vectors are the next generation of promoter reporters. With these reporters systems, you can compensate effectively for reporter background without compromising your assay’s signal intensity.

Promoterless (Traditional)

Monitor transcription from different promoters or promoter/enhancer combinations inserted into the MCS located upstream of the Living Colors fluorescent protein coding sequence.

Recombinant Fluorescent Proteins—GFP, DsRed

These Living Colors recombinant proteins are purified from E. coli. Each recombinant variant has absorption and emission spectra identical to those for the expressed proteins.

  • Purified, recombinant proteins from E. coli
  • Protein retains the absorption and emission spectra identical to expressed proteins
  • Retains fluorescence capability

Organelle and Subcellular Labeling

Visualize subcellular structures directly and noninvasively by fluorescence microscopy.

  • Localize fluorescence to specific organelles or structures in living cells
  • Visualize biological processes as they occur
  • Dual- or triple-label cells with different fluorescent proteins

Transfection Control

The Living Colors Constitutive Fluorescent Protein Expression Vectors can be used as transfection markers and to label living cells.

  • Bright fluorescent proteins for labeling
  • Choice of CMV or EF1-a promoters
