1) Bicistronic IRES Vectors

IRES-containing bicistronic vectors allow the simultaneous expression of two proteins separately but from the same RNA transcript 

  • Translate your gene of interest and an antibiotic resistance marker or fluorescent protein from a single mRNA
  • Quickly identify cells expressing your protein of interest at high levels
  • Bicistronic expression allows faster, better stable clone selection

Applications

  • Rapidly select stable clones that show high-level expression of your target protein
  • Identify transfected cells by antibiotic selection, fluorescence microscopy, or flow cytometry

 

2) Bidirectional Promoter Vectors

The pBI bidirectional vectors are designed to constitutively express a protein of interest and a reporter protein (or two proteins of interest). Expression is driven by constitutively active bidirectional human cytomegalovirus promoters (PminCMV). The gene(s) of interest must contain an initiation codon and a stop codon.

 

  • To generate stable clones using these bidirectional vectors it is recommend cotransfection with a linearized selection marker or a vector containing an antibiotic selection marker
  • Applications - Constitutive expression of a protein of interest and a reporter protein
  • You can choose from green or red fluorescent reporters

 

  • pBI-CMV1
    • Allows the constitutive expression of two proteins of interest
  • pBI-CMV2
    • Allows you to express a protein of interest and AcGFP1
    • Monomeric green fluorescent protein derived from Aequorea coerulescens
  • pBI-CMV3 allows you to express a protein of interest
    • ZsGreen1, an extremely bright green fluorescent protein
    • Derived from Zoanthus sp. reef coral
  • pBI-CMV4
    • Allows you to express a protein of interest
    • DsRed2, a red fluorescent protein derived from Discosoma sp. reef coral
  • pBI-CMV5
    • Allows you to express a protein of interest and firefly luciferase
    • It is sold as part of the Bidirectional Secreted Luciferase System, together with its reporter assay
