iDimerize Systems

All essential processes of the cell are regulated by a complex network of protein-protein interactions. iDimerize technology allows you to induce the interaction between two proteins, or the re-localization of a protein, and thereby control any cellular process that is activated (or inactivated) by these events.

 

 

1) iDimerize Inducible Homodimer System

  • System brings protein-protein interactions ubnder real-time, small molecule control
  •  A protein of interest is fused to the DmrB binding domain
  • Dimerization is induced by adding the B/B Homodimerizer
  • Rapid, chemical induction of cell signaling pathways
  • Activate any pathway or cellular event controlled by homooligomers (e.g. dimers, trimers, or tetramers)

 

2) iDimerize Inducible Heterodimer System

  • System can be used to create and control specific interactions between two different proteins
  • Achieve small-molecule control of cellular events that are usually activated by protein heterodimerization
  • Control the localization of a target protein

 

 

3) iDimerize Regulated Transcription System

Transcription factors are bifunctional proteins that recognize specific DNA sequences near target genes (via the DNA binding domain) and then recruit the transcriptional machinery of the cell to activate transcription.

  • Tightly regulated transcription using iDimerize technology
  • Dose-dependent, small molecule control of transcription
  • Activation domain is physically separated from the DNA prior to addition of the dimerizer ligand, resulting in low background

 

4) iDimerize Reverse Dimerization System (Inducible Secretion)

  • Brings the disruption of protein complexes under real-time, small molecule control
  • A protein of interest is fused to the DmrD binding domain
  • The fusion protein molecules aggregate unless the D/D Solubilizer ligand is present
  • Inducible secretion of proteins
  • Inducible dissociation/disaggregation of proteins

 

 

5) Ligands for Chemically Induced Dimerization (CID)

  • The B/B Homodimerizer (AP20187) is a synthetic, cell-permeable ligand
  • Ligand can be used to induce homodimerization of fusion proteins containing the DmrB domain
  • Small molecules used for chemical-induced dimerization 

 

6) iDimerize Antibodies

After fusing your protein of interest to one to the dimerization tags provided with iDimerize Homodimer Systems (DmrB) or iDimerize Heterodimer Systems (DmrA and DmrC), you may wish to montitor the protein's expression by SDS-PAGE followed by Western blot.

  • Detection of DmrB domains used for iDimerize Homodimer Systems 
  • Detection of DmrA and DmrC domains used for iDimerize Heterodimer Systems

 

7) Using iDimerize Inducible Protein Dimerization In Vivo

  • The B/B Homodimerizer (AP20187) has been the most widely used in vivo, and typically investigators have used doses of 0.5–10 mg per kg for each injection
  • Small molecules used for chemical-induced dimerization (CID)
  • Larger 5 mg and 25 mg sizes for in vivo use
