Tet-On and Tet-Off 1st Generation

For more than 15 years, Clontech’s Tet-On and Tet-Off brands have been the highest-performing and most-cited mammalian expression systems. During this time, a series of improvements to the system have resulted in three distinct generations of the technology

 

Tet-On and Tet-Off 2ndGeneration

Compared to the original Tet-On and Tet-Off Systems, our Tet-Advanced Systems have been optimized for improved expression in mammalian cells by utilizing human codon preferences and removing cryptic splice sites from the mRNA sequence of the transactivator.

 

Tet-tTS Transcriptional Silencer

pTet-tTS expresses the tetracycline/doxycycline-controlled transcription silencer (tTS), which is a fusion of the Tet Repressor Protein (TetR) with a KRAB silencing domain. tTS can be used to significantly lower the basal expression from 1st generation Tet-On Inducible Expression Systems.

Tetracycline-Inducible Expression-Tet-On Cell Lines

Tet-On cell lines are host cell lines that greatly simplify the task of setting up a tetracycline-inducible gene expression system that is activated by adding doxycycline (Dox) to the culture medium. Tet-On 3G cell lines, which are our highest performing pre-made cell lines for this system, are located on a separate page.

 

Tetracycline-Inducible Expression-Tet-Off Cell Lines

Tet-Off and Tet-Off Advanced inducible cell lines are host cell lines that greatly simplify the task of setting up tetracycline-inducible gene expression systems which are activated by removing doxycycline (Dox) from the culture medium. Our Tet-On Advanced and Tet-On inducible cell lines are also available, which are activated by adding Dox to the culture medium.
